In vitro maturation of affinity of CD antibodies by CHO cell display

  • Cluster of Differentiation (CD) markers serve as critical tools for immune cell classification in flow cytometry. However, domestic diagnostic antibody manufacturers in China have long been constrained by reagent quality limitations, which hinder their competitiveness against wellestablished international suppliers. To tackle this bottleneck, we employed a Chinese hamster ovary (CHO) cell display-based in vitro evolution system to optimize the enhanced cellular detection and stability of antibodies targeting CD33 and CD56. Compared with the parental antibody clones, the resultant mutant clones exhibited significantly enhanced sensitivity in detecting peripheral blood leukocytes, alongside improved stability. This study provides a feasible strategy for elevating the quality of CD-specific antibodies, with the potential to bolster high-quality domestic antibody production and advance diagnostic performance across a wide spectrum of CD markers.
